方案1：抗小鼠CD3单克隆抗体固定方法
1. 小鼠T细胞生长培养基营养系统：
- RPMI 1640培养基，ATCC改良型（含L-谷氨酰胺、丙酮酸钠和HEPES）
- 12%优级胎牛血清
- 1%抗生素-抗霉菌剂
- 细胞冻存：免疫细胞无血清冻存培养基
- 注意：上述培养基系统仅供参考。
2. 将NA/LE Rat anti-mouse CD3 Recombinant mAb稀释至PBS中，最终浓度为5µg/mL。
3. 将稀释后的NA/LE Rat anti-mouse CD3 Recombinant mAb按每孔1mL加入24孔板中固定。
4. 在37°C、5% CO₂条件下孵育2小时，或在4°C下孵育过夜。
5. 从24孔板中移除NA/LE Rat anti-mouse CD3 Recombinant mAb。
6. 准备小鼠脾脏的单细胞悬液。对于T细胞激活，细胞应重悬于完全培养基中，浓度为(1-3)×10⁶细胞/mL。对于24孔板，推荐浓度为1×10⁶细胞/mL。
7. 在V型管中准备完全补充的T细胞培养基，向步骤6中的细胞悬液中加入以下细胞因子和抗体：
- 5µg/mL NA/LE Syrian Hamster anti-mouse CD28 mAb
- 10ng/mL IL-2 Protein, Mouse
8. 将所需量的细胞转移到合适的细胞培养板中，最终密度为(1-1.5)×10⁶细胞/mL/cm²。
9. 在37°C、5% CO₂条件下孵育2天。
10. 培养2天后，检查细胞的健康状况并记录图像。
11. 以400×g离心细胞悬液5分钟，弃去上清液。用PBS洗涤细胞沉淀。
12. 以400×g离心细胞悬液5分钟，弃去上清液。将细胞重悬于0.5mL 1%牛血清白蛋白（BSA）中，计数细胞。检测细胞活性，要求活性>95%。
13. 用10µg小鼠IgG抗体阻断1×10⁶细胞。在冰浴中孵育30分钟。
14. 根据说明书加入FITC大鼠抗小鼠CD25抗体（S-R441）（S0B5215），在4°C下孵育30分钟。
15. 以400×g离心细胞悬液5分钟，用含1% BSA的PBS洗涤细胞沉淀两次。
16. 将细胞沉淀重悬于200µl PBS中，用于流式细胞术分析（需>10000个细胞）。
