  • CREB (cAMP response element-binding protein) is a well-known transcription factor that plays a pivotal role in the regulation of gene expression in response to various stimuli, including neuronal activity, hormones, and growth factors. Phosphorylation of CREB at serine 133 (Ser133) is a critical event for its activation, as it enhances the binding of CREB to the cAMP response element (CRE), leading to the transcription of target genes involved in processes such as learning, memory, and cell survival. The phosphorylation of CREB at Ser133 is typically catalyzed by various protein kinases, including protein kinase A (PKA), calcium/calmodulin-dependent protein kinase IV (CaMKIV), and mitogen-activated protein kinase (MAPK). This phosphorylation event is a robust indicator of neuronal activation and synaptic plasticity, making it a widely studied marker in neurobiological research.

  • ChIP

    • Chromatin immunoprecipitation (ChIP) was performed on HeLa cells cross - linked with 1% formaldehyde for 10 min, then chromatin was fragmented by sonication. Parallel reactionsused CREB (phospho S133) Recombinant Rabbit mAb (S-1100-102) and IgG IsotypeControl (SDT-R173) at 1:50 for immunoprecipitation.
      Post-immunoprecipitation, both samples were washed, eluted, and cross - links reversed. Purified DNA was analyzed by qPCR.

      qPCR showed the enrichment of ALS2, NR4A3 and SAT-α in
      CREB (phospho S133) Recombinant Rabbit mAb (S-1100-102)-immunoprecipitated
      sample.

我们推荐客户使用TPST+5%脱脂奶粉来稀释一抗,进行封闭。 虽然BSA被推荐为WB检测磷酸化蛋白的常用封闭剂,但是脱脂奶粉获取更加方便,覆盖更广泛的非特异性结合位点,在一抗性能优越的前提下,使用脱脂奶粉封闭性价比更高
